產(chǎn)品介紹 This gene encodes a homodimeric transmembrane protein which is a major glycoprotein of the vascular endothelium. This protein is a component of the transforming growth factor beta receptor complex and it binds to the beta1 and beta3 peptides with high affinity. Mutations in this gene cause hereditary hemorrhagic telangiectasia, also known as Osler-Rendu-Weber syndrome 1, an autosomal dominant multisystemic vascular dysplasia. This gene may also be involved in preeclampsia and several types of cancer. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. Function:
Major glycoprotein of vascular endothelium. May play a critical role in the binding of endothelial cells to integrins and/or other RGD receptors.

Subunit:
Homodimer that forms an heteromeric complex with the signaling receptors for transforming growth factor-beta: TGFBR1 and/or TGFBR2. It is able to bind TGF-beta 1, and 3 efficiently and TGF-beta 2 less efficiently. Interacts with TCTEX1D4. Interacts with ARRB2.

Subcellular Location:
Membrane; Single-pass type I membrane protein.

Tissue Specificity:
Endoglin is restricted to endothelial cells in all tissues except bone marrow.

DISEASE:
Defects in ENG are the cause of hereditary hemorrhagic telangiectasia type 1 (HHT1) [MIM:187300]; also known as Osler-Rendu-Weber syndrome 1 (ORW1). HHT1 is an autosomal dominant multisystemic vascular dysplasia, characterized by recurrent epistaxis, muco-cutaneous telangiectases, gastro-intestinal hemorrhage, and pulmonary (PAVM), cerebral (CAVM) and hepatic arteriovenous malformations; all secondary manifestations of the underlying vascular dysplasia. Although the first symptom of HHT1 in children is generally nose bleed, there is an important clinical heterogeneity.

CD105(Endoglin):CD105是一種分子量為180kDa的存在于細(xì)胞表面的同源二聚體跨膜糖蛋白,是TGF-β受體復(fù)合物的組成部分,是TGF-β的附屬受體,能與多種TGF-β超家族成員結(jié)合尤其與TGF-β1、TGF-β3有很高的親和力,調(diào)節(jié)TGF-βs與其受體結(jié)合而參與信號(hào)傳導(dǎo),是內(nèi)皮細(xì)胞增殖相關(guān)膜抗原,在培養(yǎng)的高增殖活性內(nèi)皮細(xì)胞和許多惡性腫瘤組織血管內(nèi)皮細(xì)胞中高表達(dá),參與血管生成,但其在血管生成調(diào)節(jié)中的作用機(jī)制尚未闡明。CD105基因定位于人9號(hào)染色體,是Ⅰ型遺傳性出血性毛細(xì)血管擴(kuò)張癥的相關(guān)基因。CD105基因敲除小鼠胚胎因血管生成缺陷而于受精后平均11.5天死亡,故有學(xué)者認(rèn)為,CD105可能與血管生成啟動(dòng)有關(guān),可用于標(biāo)記腫瘤新生血管。 Endoglin是內(nèi)皮細(xì)胞表面與細(xì)胞增殖相關(guān)的膜抗原,也是轉(zhuǎn)化生長(zhǎng)因子β超家族受體復(fù)合物成分之一。其具有調(diào)節(jié)內(nèi)皮細(xì)胞對(duì)TGF的反應(yīng)、促內(nèi)皮細(xì)胞增殖和促血管形成等功能,與腫瘤血管的發(fā)生密切相關(guān)。近年來,Endoglin在腫瘤診斷、判斷預(yù)后和療效及抗腫瘤血管靶向治療等方面的作用得到重視。姬,2017.1.15;
